چکیده

Background A thorough knowledge of the metabolism and its compartmentation in mammalian cells is desirable to enable rational design and optimization of producing cell lines and production processes for biopharmaceuticals. In this study we focused on acquiring a detailed understanding of metabolite channelling and the metabolic flux distribution during overflow metabolism in the human cell line AGE1.HN (ProBioGen AG). This metabolic phenotype characterized by energy spilling as well as waste product formation is commonly observed in the beginning of the cultivation [1]. C tracer experiments and C flux analysis as applied in this investigation represent methods offering in-depth insights into cellular physiology [2,3].

Producer vs. parental cell – metabolic changes and burden upon α1-antitrypsin production in AGE1.HN®

Background The human designer cell line AGE1.HN represents a promising production system for biopharmaceuticals, particularly for those needing human-type post-translational modifications [1,2]. For further rational improvement of the cell line and the cultivation process a detailed understanding of the metabolism and metabolic changes during glycoprotein production is desirable. Metabolism and...

Cytotoxic and antimicrobial effects of lactobacillus sakei on human colorectal adenocarcinoma cell line (HT29) and some pathogenic microorganisms

Background: Even after surgery, as the most effective treatment for colorectal cancer, about 30-40% of cases are recurring. Since growth inhibition is an important strategy in cancer treatment, many attempts are in the program to find new agents, so in this study, the cytotoxic and antimicrobial effects of Lactobacillus sakei on colon cancer cell line (HT-29) and some pathogenic microorganisms ...

Analysis of the mitochondrial subproteome of the human cell line AGE1.HN – a contribution to a systems biology approach

Background In Systems Biology approaches a good amount of reliable data is essential for effective modelling of distinct biological processes. Hence the focus of the SysLogicsProject was on modelling of the central metabolism using different functional genomic techniques. For displaying the involved proteins, it is crucial to enrich them using subproteomic fractions. As many enzymes being invol...

Quantitative characterization of metabolism and metabolic shifts during growth of the new human cell line AGE1.HN using time resolved metabolic flux analysis

For the improved production of vaccines and therapeutic proteins, a detailed understanding of the metabolic dynamics during batch or fed-batch production is requested. To study the new human cell line AGE1.HN, a flexible metabolic flux analysis method was developed that is considering dynamic changes in growth and metabolism during cultivation. This method comprises analysis of formation of cel...

Study of crocin & radiotherapy-induced cytotoxicity and apoptosis in the head and neck cancer (HN-5) cell line

Malignant tumors of head and neck carcinomas are the sixth most common type of cancer. Current systemic therapies for cancer show side effects in normal tissues and short-term efficacy due to drug resistance. Consequently, there is much interest in identifying new drugs for cancer treatment. Crocin (an active ingredient of saffron) has been shown to have cytotoxic effects on cancer cell lines. ...
